How exactly do I host a server with Windows, a friend and I want to have a private jam. Any help would be appreciated. :)

Provided your windows machine has adequate bandwidth, running a server on it is no problem. The steps:
Download the win32 server: http://www.ninjam.com/downloads/ninjam_server_win32.zip
extract the zip to a directory, edit example.cfg to your liking, then open a command prompt, cd to that directory, and run:
ninjamserver example.cfg

Cool, then what does client type on their end to access my server? Is it ip & port like, 00.00.00.00:2049?

yes, they can use the IP of your server followed by colon port, or the hostname of your server followed by colon port, just like you have.
