Now is impossible. In this case you need click in the general REAPER interface and later use marker´s shortcut. After this, you must go back to the plugin in order to continue adjusting it.
It's not impossible. If you assign any keyboard shortcut that includes a
CTRL (CMD) or ALT (OPT) modifier, eg. CTRL+M to the action, it will not
be 'eaten' by the plug-in (unlike solo keys and shift-key commands).
Also you can assign MIDI, which will always work (regardless of focus), as
long as the reaper is processing (there are settings to stop running while
not in focus).
