USB cable - the one that comes with the Scarlett is a cheap one, so might be knackered.
Check you're getting signal at the Focusrite Mix Control software.
If not, make sure you've got Mix Control set properly.
If you are, you've got Reaper setup wrong.
If Reaper is set properly, Mix Control is set properly, a new cable didn't fix it, the Scarlett's got an internal problem.
NB: If you've veered away from defaults settings in either program, it may be worth doing a 'portable' install of Reaper - a tickbox at the install process that allows you to install a completely independent install to a single folder - and a reset to default of the Mix Control software, for test purposes and/or maybe even reinstall the FR driver.
