In the short answer when you go to Preferances/Project theres a
location field/browser button above it.
In this field you have to point Reaper to a template file that you wish it to open as a default project.

I was gonna say the same thing. You may have
Load template project when starting up and not loading last project ticked, but you indeed need an actual template project file for it to work.
To do this, start a new project then save it somewhere - I saved mine in the Reaper directory. Next, point Reaper to that file as the above post describes.
